I believe unless you use a very unusual seal system you will have difficulty holding pressure cannister. Most seals are lipped seals and can only hold significant pressure from one direction.
You could use two seals with one facing each way to accomplish your goal. I am going to use a standard seal arrangement and usi a first stage scuba regulator to maintain a similar pressure inside with relationship to whats outside.
 
I have not built mine yet, so I am sure others have more specific info and possibly ideas.
