I took a look in one of my books. It depends on what you call "comfortable" but there were a lot of two-man subs build with only a 1500 mm (5 feet) spherical pressurehull (Deep Jeep, Benthos, Star II). Alvin has a seven foot sphere and occupies 3 men. Even a 2000 mm (6 foot 8)  sphere is big enough for 3 men (Pisces I, Deepstar 4000).

I am currently in the process of designing (no hopes yet of building, only design) a small two-man sub called the Manta, taking its name from the design similarity to the manta ray. My question is: would a seven foot diameter sphere be large enough to seat two people with a control system comfortably? (I'm quite sure five feet would be too small, but I don't envision spheres very well–they're such an awkward shape, lol)
